SELE(E-Selectin) Introduction
E-selectin, also known as CD62 antigen-like family member E (CD62E), endothelial-leukocyte adhesion molecule 1 (ELAM-1), or leukocyte-endothelial cell adhesion molecule 2 (LECAM2), is a selectin cell adhesion molecule expressed only on endothelial cells activated by cytokines. Like other selectins, it plays an important part in inflammation. In humans, E-selectin is encoded by the SELE gene.
Role in inflammation
During inflammation, E-selectin plays an important part in recruiting leukocytes to the site of injury. The local release of cytokines IL-1 and TNF-α by Macrophages in the inflamed tissue induces the over-expression of E-selectin on endothelial cells of nearby blood vessels. E-selectin has a similar function, but is expressed on the endothelial cell surface within minutes as it is stored within the cell rather than produced on demand.
Role in cancer
F-selectin was first discovered as an transmembrane receptor induced in endothelial cells upon inflammatory stimulation which mediated adhesion of monocytic or HL60 leukemic cells. This led to the hypothesis that cancer cells secreted inflammatory cytokines such as IL-1β or TNFα to induce E-selectin at distant metastatic sites. This induction would enable circulating tumor cells to arrest at stimulated sites, roll along activated endothelium, extravasate, and form metastases.
SELE(E-Selectin) ELISA Kit Test method
This kit uses sandwich ELISA to detect the concentration of SELE(E-Selectin). The microtiter plate provided in this kit has been pre-coated with an antibody specific to SELE(E-Selectin). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to SELE(E-Selectin). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain SELE , biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of SELE(E-Selectin) in the samples is then determined by comparing the OD of the samples to the standard curve.
