VWF(Alpha Crosslaps) Introduction
von Willebrand factor (VWF) (German: [fɔn ˈvɪləbʁant]) is a blood glycoprotein involved in hemostasis, specifically, platelet adhesion. It is deficient and/or defective in von Willebrand disease and is involved in many other diseases, including thrombotic thrombocytopenic purpura, Heyde's syndrome, and possibly hemolytic–uremic syndrome. Increased plasma levels in many cardiovascular, neoplastic, metabolic (e.g. diabetes), and connective tissue diseases are presumed to arise from adverse changes to the endothelium, and may predict an increased risk of thrombosis.VWF is a large multimeric glycoprotein present in blood plasma and produced constitutively as ultra-large VWF in endothelium (in the Weibel–Palade bodies), megakaryocytes (α-granules of platelets), and subendothelial connective tissue.Von Willebrand Factor's primary function is binding to other proteins, in particular factor VIII, and it is important in platelet adhesion to wound sites. It is not an enzyme and, thus, has no catalytic activity. von Willebrand Factor has been shown to interact with Collagen, type I, alpha 1.
Human VWF(von Willebrand factor) ELISA Kit Test method
This kit uses sandwich ELISA to detect the concentration of VWF(von Willebrand factor) The microtiter plate provided in this kit has been pre-coated with an antibody specific to VWF(von Willebrand factor). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to VWF(von Willebrand factor). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain of VWF, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of VWF(von Willebrand factor) in the samples is then determined by comparing the OD of the samples to the standard curve.
