PTGS2/COX-2(Prostaglandin G/H synthase 2) Introduction
Cyclooxygenase (COX), officially known as prostaglandin-endoperoxide synthase (PTGS), is an enzyme (specifically, a family of isozymes, EC 1.14.99.1) that is responsible for formation of prostanoids, including thromboxane and prostaglandins such as prostacyclin, from arachidonic acid. A member of the animal-type heme peroxidase family, it is also known as prostaglandin G/H synthase. The specific reaction catalyzed is the conversion from arachidonic acid to Prostaglandin H2, via a short-living Prostaglandin G2 intermediate. Inhibition of COX-2 using celecoxib has been shown to reduce the immunosuppressive TGFβ expression in hepatocytes attenuating EMT in human hepatocellular carcinoma.
Rat PTGS2/COX-2(Prostaglandin G/H synthase 2) ELISA Kit Test method
This kit uses sandwich ELISA to detect the concentration of PTGS2/COX-2(Prostaglandin G/H synthase 2). The microtiter plate provided in this kit has been pre-coated with an antibody specific to PTGS2/COX-2(Prostaglandin G/H synthase 2). Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to PTGS2/COX-2(Prostaglandin G/H synthase 2). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain PTGS2/COX-2(Prostaglandin G/H synthase 2), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of PTGS2/COX-2(Prostaglandin G/H synthase 2) in the samples is then determined by comparing the OD of the samples to the standard curve.
