TJP1(Tight junction protein ZO-1) Introduction

Zonula occludens-1 ZO-1, also known as Tight junction protein-1 is a 220-kD peripheral membrane protein that is encoded by the TJP1 gene in humans. It belongs to the family of zonula occludens proteins (ZO-1, ZO-2, and ZO-3), which are tight junction-associated proteins and of which, ZO-1 is the first to be cloned.This gene encodes a protein located on a cytoplasmic membrane surface of intercellular tight junctions. The encoded protein may be involved in signal transduction at cell–cell junctions. Two transcript variants encoding distinct isoforms have been identified for this gene.Tight junction protein 1 has been shown to interact with: F11 receptor, GJA3, GJA8, Gap junction protein, alpha 1, KIRREL, MLLT4, Occludin, TJP3, and Tight junction protein 2.

Human TJP1(Tight junction protein ZO-1) ELISA Kit Test method

This kit uses sandwich ELISA to detect the concentration of TJP1(Tight junction protein ZO-1) The microtiter plate provided in this kit has been pre-coated with an antibody specific to TJP1(Tight junction protein ZO-1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to TJP1(Tight junction protein ZO-1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain of TJP1(Tight junction protein ZO-1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of TJP1(Tight junction protein ZO-1) in the samples is then determined by comparing the OD of the samples to the standard curve.
