Detection principle
Reduced GSH can react with Dinitrobenzoic acid (DNTB) to form a yellow complex which can be detected by colorimetric assay at 405 nm and calculate the reduced GSH content indirectly.

The level of GSH was significantly lower (P<0.05) in DMH treated groups (G3-G6) comparing to control group (G1). GSH level was significantly increased (P<0.05) in quercetin treated groups (G4-G6). Quercetin alone in healthy controls did not have any impact on the GSH level (G2).
S G Darband et al found that quercetin attenuated oxidative DNA damage in rats with DMH induced colon carcinogenesis, changes of antioxidants can be obtained. Reduced glutathione level of rat colon tissue was determined using GSH colorimetric assay kit (E-BC-K030-M).
Dilution of sample
It is recommended to take 2~3 samples with expected large difference to do pre-experiment before formal experiment and dilute the sample according to the result of the pre-experiment and the detection range (2-100 μmol/L).
