Description

This Denosumab ELISA is for determination of Denosumab in human serum, plasma and cell culture supernatant. Denosumab is a human monoclonal antibody used for the treatment of osteoporosis, treatment-induced bone loss, metastases to bone, and giant cell tumor of bone. It is sold under the trade name Prolia or Xgeva. Denosumab works by lowering the hormonal message that leads to excessive osteoclast-driven bone removal. An Ig2 mAb, Denosumab is a RANK ligand inhibitor. It prevents development of osteoclasts, which are cells that cause bone resorption. Denosumab protects bone from degradation, and helps to counter the progression of the disease.The method employs the quantitative sandwich enzyme immunoassay technique. Antibodies to Denosumab are pre-coated onto microwells. Samples and standards are pipetted into microwells and human Denosumab present in the sample are bound by the capture antibody. Then, a HRP (horseradish peroxidase) conjugated anti- Denosumab antibody is pipetted and incubated. After washing microwells in order to remove any non-specific binding, the ready to use substrate solution (TMB) is added to microwells and color develops proportionally to the amount of Denosumab in the sample. Color development is then stopped by addition of stop solution. Absorbance is measured at 450 nm.
