TIMP-1(Metalloproteinase inhibitor 1) Introduction

TIMP metallopeptidase inhibitor 1, also known as TIMP1, a tissue inhibitor of metalloproteinases, is a glycoprotein with a molecular weight of 28 kDa. TIMP1 is expressed from several tissues of organisms.This protein is a member of the TIMP family. The glycoprotein is a natural inhibitor of the matrix metalloproteinases (MMPs), a group of peptidases involved in degradation of the extracellular matrix. In addition to its inhibitory role against most of the known MMPs, the encoded protein is able to promote cell proliferation in a wide range of cell types, and may also have an anti-apoptotic function.TIMP1 is an inhibitory molecule that regulates matrix metalloproteinases (MMPs), and disintegrin-metalloproteinases (ADAMs and ADAMTSs). In regulating MMPs, TIMP1 plays a crucial role in extracellular matrix (ECM) composition, wound healing, and pregnancy.

Human TIMP-1(Metalloproteinase inhibitor 1) ELISA Kit Test method

This kit uses sandwich ELISA to detect the concentration of TIMP-1(Metalloproteinase inhibitor 1) The microtiter plate provided in this kit has been pre-coated with an antibody specific to TIMP-1(Metalloproteinase inhibitor 1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to TIMP-1(Metalloproteinase inhibitor 1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain of TIMP-1(Metalloproteinase inhibitor 1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of TIMP-1(Metalloproteinase inhibitor 1) in the samples is then determined by comparing the OD of the samples to the standard curve.
