Description

DKW Basal Salts With Macronutrients and Micronutrients is a powdered plant tissue culture medium based on the Driver and Kuniyuki (DKW) formulation, as described by Driver and Kuniyuki (1984) and McGranahan et al. (1987).

The formulation provides macronutrients and micronutrients required for controlled in vitro plant culture, plant tissue culture and micropropagation. DKW medium is particularly associated with the culture and regeneration of woody plants, including walnut, and can be used in workflows involving multiple shoot induction, callus-mediated shoot regeneration and plant propagation.

DKW Medium Preparation Protocol

The supplied protocol describes preparation of 1 liter of DKW tissue culture medium as follows:

  1. Add 800 mL of distilled water to a suitable beaker.
  2. Add 5.36 g of DKW media, 30 g of sugar and PPM™ according to the supplied protocol, generally 1–2 mL/L.
  3. Make up the volume to 1000 mL with water.
  4. Adjust the pH according to the tissue culture protocol; the supplied preparation specifies a range of 5.4–5.8.
  5. Add a suitable gelling agent, such as agar at 6–8 g/L or gellan gum at 3–4 g/L, while stirring.
  6. Autoclave the medium for 20 minutes at 15 psi and 121°C.
The prepared medium is ready for tissue culture use after appropriate sterilization and cooling.

Storage Conditions

Dehydrated plant tissue culture media is hygroscopic and should be protected from moisture and direct sunlight. Store the powder at room temperature unless otherwise specified on the product label.

Once prepared, the medium should be stored at 2–8°C away from direct light and used before the stated expiry date.

After opening, the unused dehydrated medium should be protected from moisture. The supplied storage instructions recommend using the contents promptly or storing the unused portion in a desiccator and under refrigeration at 2–8°C.
