Description

Kinetin, also known as 6-furfurylaminopurine, is a synthetic cytokinin used in plant tissue culture research. Cytokinins influence cell division, shoot development, and several aspects of plant growth and development.

When incorporated into suitable growth media, such as Murashige and Skoog (MS) medium, kinetin is commonly used in combination with auxins to regulate plant cell differentiation and organ development.

CAS Number: 525-79-1

Applications

  • Shoot induction and proliferation in plant tissue culture.
  • Support of cell division in suitable culture systems.
  • Callus development and shoot initiation, depending on the plant species and culture conditions.
  • Use in combination with auxins to adjust the hormonal balance and influence root and shoot development.
  • Research into bud development, tissue growth, and plant senescence.

Assay Protocol

General guidance for preparing plant tissue culture medium supplemented with kinetin:

  1. Prepare the selected basal medium, such as Murashige and Skoog (MS) or DKW medium, according to the intended application.
  2. Add a suitable carbohydrate source. A commonly used starting point is 30 g/L sucrose, subject to the culture protocol.
  3. If required, add a validated gelling agent, such as approximately 3 - 4 g/L gellan gum or 6 - 8 g/L agar. Follow the medium manufacturer's instructions.
  4. Prepare a kinetin stock solution using a suitable solvent. Kinetin is very slightly soluble in water, so select the solvent and preparation method appropriate for the application.
  5. Add the required volume of stock solution to reach the target final kinetin concentration. Adjust the concentration according to the plant species and experimental objective.
  6. Adjust the medium pH and sterilize using a validated procedure suitable for the complete formulation. If needed, prepare and sterilize the kinetin stock separately using a compatible method before aseptic addition.
Important: Solubility, solvent compatibility, sterilization conditions, and optimal kinetin concentration depend on the formulation and plant species. Confirm these parameters with your validated laboratory protocol.
