Description

Detection principle

Fe3+ can be deoxidized to Fe2+ by VE with ferroin existing. Fe2+ can react with phenanthroline and form pink compound under certain condition. After colorimetric assay, VE content can be figured out according to the standard curve or calculated through formula.

The level of vitamin E was significantly lower (P<0.05) in DMH treated groups (G3) comparing to control group (G1). VE level was not significantly changed in initiation phase treatment group (G4), but resulted in significant enhancement (P<0.05) in post-initiation (G5) and entire phase treatment group (G6). Quercetin alone in healthy controls did not have any impact on the VE level (G2).

S G Darband et al found that quercetin attenuated oxidative DNA damage in rats with DMH induced colon carcinogenesis, changes of antioxidants can be obtained. Vitamin E level of rat colon tissue was determined using VE colorimetric assay kit (E-BC-K033-M).

Dilution of sample

It is recommended to take 2~3 samples with expected large difference to do pre-experiment before formal experiment and dilute the sample according to the result of the pre-experiment and the detection range (0.95-40 μg/mL).
